Ber-H2 (CD30) Immunohistochemical Staining of Human Fetal Tissues
OBJECTIVE: CD30 antigen has long been considered to be restricted to the tumor cells of Hodgkin's disease and of anaplastic large cell lymphoma as well as to T and B activated lymphocytes. It is now apparent that the range of normal and neoplastic cells, which may express CD30 antigen, is much wider than what at first thought. In order to gain insight into the physiological function of CD30 antigen, we studied the distribution of its expression in the tissues of fetuses from week 8th to week 16th. MATERIALS AND METHODS: We investigated the immunohistochemical expression of CD30 antigen in paraffin-embedded tissue samples representing all systems from 30 fetuses after therapeutic abortion at 8th to 10th and 12th to 16th week of gestation, respectively, using the monoclonal antibody Ber-H2. RESULTS: Our results demonstrated that CD30 is expressed early in human fetal development (8th to 10th week of gestation) in several fetal tissues derived from all three germ layers (gastrointestinal tract, special glands of the postpharyngeal foregut, urinary, musculoskeletal, reproductive, nervous , endocrine systems), with the exception of the skin and hematolymphoid system (thymus), in which the antigen is expressed later on (10th week onwards). Expression of CD30 was restricted to the hematolymphoid system in the 12-16 weeks of gestation. No expression of the marker was observed in the respiratory and cardiovascular systems during the entire period examined. CONCLUSIONS: CD30 antigen is of importance in cell development, and proliferation. It is also pathway-related to terminal differentiation in many fetal tissues and organs. PMID: 16244703

Base lesions in DNA can stall the replication machinery or induce mutations if bypassed. Consequently, lesions must be repaired before replication or in a post-replicative process to maintain genomic stability. Base excision repair (BER) is the main pathway for repair of base lesions and is known to be associated with DNA replication, but how BER is organized during replication is unclear. Here we coupled the iPOND (isolation of proteins on nascent DNA) technique with targeted mass-spectrometry analysis, which enabled us to detect all proteins required for BER on nascent DNA and to monitor their spatiotemporal orchestration at replication forks. We demonstrate that XRCC1 and other BER / single-strand break repair (SSBR) proteins are enriched in replisomes in unstressed cells, supporting a cellular capacity of post-replicative BER / SSBR. Importantly, we identify for the first time the DNA glycosylases MYH, UNG2, MPG, NTH1, NEIL1, 2 and 3 on nascent DNA. Our findings suggest that a broad spectrum of DNA base lesions are recognized and repaired by BER in a post-replicative process. © The Author (s) 2017. A large number of diverse, complex, and distributed data resources are currently available in the Bioinformatics domain. The pace of discovery and the diversity of information means that centralized reference databases like UniProt and Ensembl cannot integrate all potentially relevant information sources. From a user perspective, however, centralized access to all relevant information concerning a specific query is essential. The Distributed Annotation System (DAS) defines a communication protocol to exchange annotations on genomic and protein sequences; This standardization enables clients to retrieve data from a myriad of sources, thus offering centralized access to end users. We introduce MyDas, a web server that facilitates the publishing of biological annotations according to the DAS specification. It deals with the common functionality requirements of making data available, while also providing an extension mechanism in order to implement the specifics of data store interaction. MyDas allows the user to define where the required information is located along with its structure, and is then responsible for the communication protocol details. PMID: 23028496

Average BER analysis of SCM-based free-space optical systems by considering the effect of IM3 with OSSB signals under turbulence channels.

In this paper, we derive the average bit error rate (BER) of subcarrier multiplexing (SCM) -based free space optics (FSO) systems using a dual-drive Mach-Zehnder modulator (DD-MZM) for optical single-sideband (OSSB ) signals under atmospheric turbulence channels. In particular, we consider the third-order intermodulation (IM3), a significant performance degradation factor, in the case of high input signal power systems. The derived average BER, as a function of the input signal power and the scintillation index, is employed to determine the optimum number of SCM users upon the designing FSO systems. For instance, when the user number doubles, the input signal power decreases by almost 2 dBm under the log-normal and exponential turbulence channels at a given average BER.

The present study has been carried out to evaluate the effect of a composite edible coating of 2% sodium alginate and 0.2% olive oil with combination of 1% ascorbic acid and 1% citric acid on the post harvest nutritional quality and shelf life of Ber fruit stored at 25 ± 2 ° C and 65% RH The coatings reduced the decay occurrence, weight loss, accumulation of total soluble solids (TSS) and total sugars in Ber fruit and enhanced the level of antioxidants. The delayed activity of polygalacturonase (PG), pectate lyase (PL) and pectin methyl esterase (PME) was noticed in coated fruits than that of the control fruit indicating the reduced softening and ripening process. These findings suggest that the composite edible coating tested under the current study has the potential to control decaying incidence of Ber fruit, extends its storage life and also improves its valuable nutritional characteristics.

Analytical Evaluation of the Effect of Cross-Polarization-Induced Crosstalk on the BER Performance of a PDM-QPSK Coherent Homodyne Optical Transmission System

An analytical approach is developed to find the effect of cross-polarization (XPol) -induced crosstalk on the bit error rate (BER) performance of a polarization division multiplex (PDM) quadrature phase shift keying (QPSK) optical transmission system with polarization diversity receiver . Analytical expression for the XPol-induced crosstalk and signal to crosstalk plus noise ratio (SCNR) are developed at the output of polarization diversity PDM-QPSK coherent optical homodyne receiver conditioned on a given value of mean misalignment angle. Considering Maxwellian distribution for the pdf of the misalignment angle, the average SCNR and average BER are derived. Results show that there is significant deterioration in the BER performance and power penalty due to XPol-induced crosstalk. Penalties in signal power are found to be 8.85 dB, 11.28 dB and 12.59 dB correspondingly for LO laser power of -10 dBm, -5 dBm and 0 dBm at a data rate of 100 Gbps, mean misalignment angle of 7.5 degree and BER of 10 -9 compared to the signal power without crosstalk.

Abstract Australian stiletto flies of the sister-genera Acupalpa Kröber, 1912 and Pipinnipons Winterton, 2001 (Diptera: Therevidae: Agapophytinae) are revised. Twelve new species of Acupalpa are described, while Acupalpa imitans (White, 1915), comb. n. is transferred from Pipinnipons and Acupalpa albimanis (Kröber, 1914), comb. n. is transferred from Ectinorhynchus Macquart as a senior synonym of Acupalpa pollinosa Mann. The total number of species of Acupalpa is therefore increased to 19: Acupalpa albimanis (Kröber), comb. n., Acupalpa albitarsa male, Acupalpa boharti sp. n., Acupalpa divisa (Walker), Acupalpa dolichorhyncha sp. n., Acupalpa glossa sp. n., Acupalpa imitans (White), comb. n., Acupalpa irwini Winterton, Acupalpa melanophaeos sp. n., Acupalpa miaboolya sp. n., Acupalpa minuta sp. n., Acupalpa minutoides sp. n., Acupalpa notomelas sp. n., Acupalpa novayamarna sp. n., Acupalpa rostrata Kröber, Acupalpa semirufa man, Acupalpa westralica sp. n., Acupalpa yalgoo sp. n. and Acupalpa yanchep sp. n. Integrating sequence and structural biology with DAS

Background The Distributed Annotation System (DAS) is a network protocol for exchanging biological data. It is frequently used to share annotations of genomes and protein sequence. Results Here we present several extensions to the current DAS 1.5 protocol. These provide new commands to share alignments, three dimensional molecular structure data, add the possibility for registration and discovery of DAS servers, and provide a convention how to provide different types of data plots. We present examples of web sites and applications that use the new extensions. We operate a public registry of DAS sources, which now includes entries for more than 250 distinct sources. Conclusion Our DAS extensions are essential for the management of the growing number of services and exchange of various biological data sets. In addition, the extensions allow new types of applications to be developed and scientific questions to be addressed. The registry of DAS sources is available at PMID: 17850653
Channel correlation and BER performance analysis of coherent optical communication systems with receive diversity over moderate-to-strong non-Kolmogorov turbulence.

In this paper, new expressions of the channel-correlation coefficient and its components (the large- and small-scale channel-correlation coefficients) for a plane wave are derived for a horizontal link in moderate-to-strong non-Kolmogorov turbulence using a generalized effective atmospheric spectrum which includes finite-turbulence inner and outer scales and high-wave number "bump". The closed-form expression of the average bit error rate (BER) of the coherent free-space optical communication system is derived using the derived channel-correlation coefficients and an α-μ distribution to approximate the sum of the square root of arbitrarily correlated gamma -Gamma random variables. Analytical results are provided to investigate the channel correlation and evaluate the average BER performance. The validity of the proposed approximation is illustrated by Monte Carlo simulations. This work will help with further investigation of the fading correlation in spatial diversity systems.

Non-small cell lung cancer (NSCLC) patients harboring EGFR-activating mutations initially respond to EGFR tyrosine kinase inhibitors (EGFR-TKIs) and have shown favorable outcomes. However, acquired drug resistance to EGFR-TKIs develops in almost all patients mainly due to the EGFR T790 M mutation. Here, we show that treatment with low-dose EGFR-TKI results in the emergence of the EGFR T790 M mutation and in the reduction of HSP70 protein levels in HCC827 cells. Erlotinib treatment inhibits HSP70 phosphorylation at tyrosine 41 and increases HSP70 ubiquitination, resulting in HSP70 degradation. We show that EGFR-TKI treatment causes increased DNA damage and enhanced gene mutation rates, which are secondary to the EGFR-TKI-induced reduction of HSP70 protein. Importantly, HSP70 overexpression delays the occurrence of Erlotinib-induced EGFR T790 M mutation. We further demonstrate that HSP70 interacts with multiple enzymes in the base excision repair (BER) pathway and promotes not only the efficiency but also the fidelity of BER. Collectively, our findings show that EGFR-TKI treatment facilitates gene mutation and the emergence of EGFR T790 M secondary mutation by the attenuation of BER via induction of HSP70 protein degradation. Uptake of 2S albumin allergens, Ber e 1 and Ses i 1, across human intestinal epithelial Caco-2 cell monolayers.

We have investigated the absorption rates of two purified major allergen 2S albumins, Ber e 1 from Brazil nuts (Bertholletia excelsa Humb.& Bonpl.) And Ses i 1 from white sesame seeds (Sesamum indicum L.), across human intestinal epithelial Caco-2 cell monolayers following gastrointestinal digestion in vitro. The transport from apical to basolateral side in cell monolayers was evaluated by RP-HPLC-UV and indirect competitive ELISA methods, being confirmed by western blotting analysis. Significant amounts (approximately 15-25 nmol micromol (-1) initial amount / h) of intact Ber e 1 and Ses i 1 were found in the basolateral side. The absorption rates of both plant allergens through the cell monolayer were shown to be constant during the whole incubation period (4 h at 37 degrees C), verifying that the permeability of the membrane was not altered by the allergen digests. Our findings revealed that both purified 2S albumin allergens may be able to survive in immunologically reactive forms to the simulated harsh conditions of the gastrointestinal tract to be transported across the Caco-2 cell monolayers, so that they would be able to sensitize the mucosal immune system and / or elicit an allergic response.
Nucleolar accumulation of APE1 depends on charged lysine residues that undergo acetylation upon genotoxic stress and modulate its BER activity in cells

Apurinic / apyrimidinic endonuclease 1 (APE1) is the main abasic endonuclease in the base excision repair (BER) pathway of DNA lesions caused by oxidation / alkylation in mammalian cells; within nucleoli it interacts with nucleophosmin and rRNA through N-terminal Lys residues, some of which (K27 / K31 / K32 / K35) may undergo acetylation in vivo. Here we study the functional role of these modifications during genotoxic damage and their in vivo relevance. We demonstrate that cells expressing a specific K-to-A multiple mutant are APE1 nucleolar deficient and are more resistant to genotoxic treatment than those expressing the wild type, although they show impaired proliferation. Of interest, we find that genotoxic treatment induces acetylation at these K residues. We also find that the charged status of K27 / K31 / K32 / K35 modulates acetylation at K6 / K7 residues that are known to be involved in the coordination of BER activity through a mechanism regulated by the sirtuin 1 deacetylase. Of note, structural studies show that acetylation at K27 / K31 / K32 / K35 may account for local conformational changes on APE1 protein structure. These results highlight the emerging role of acetylation of critical lys residues in regulating APE1 functions. They also suggest the existence of cross-talk between different Lys residues of APE1 occurring upon genotoxic damage, which may modulate APE1 subnuclear distribution and enzymatic activity in vivo. The astronomers' desire to penetrate deeper into space transforms into a demand for larger telescopes. The primary mirror constitutes the main part of a reflecting telescope, and it determines all subsequent activities. Already in the 1930s activities existed in the Schott company to manufacture mirror blanks up to diameters of 2 meters, which could not be pursued because of political constraints. This ambitious goal was again picked up a few years after the war. At a time when the procurement of raw materials was extremely difficult, the glass workers of Schott in Jena attacked this large project. After some failures, a good mirror blank could be delivered to the Carl Zeiss Company in 1951 for further processing and for the construction of the first 2-meter reflecting telescope. From 1960 to 1986, this mirror made from optical glass ZK7 served its purpose at the Karl Schwarzschild Observatory in Tautenburg. lt was then replaced by a zero expansion glass ceramics mirror.
